Background
The breast cancer resistance protein (BCRP/ABCG2), encoded by the ABCG2 gene, is a 655-amino-acid, 72–75 kDa half-transporter (a member of the ABC transporter superfamily G subfamily) that forms homodimers or oligomers to function as an ATP-dependent efflux pump actively transporting a broad range of substrates—including anticancer drugs (e.g., mitoxantrone, topotecan), endogenous compounds (e.g., estrone-3-sulfate, uric acid), and xenobiotics—across apical membranes of tissues such as the placenta, intestine, liver, kidney proximal tubules, and brain microvascular endothelium, thereby contributing to drug absorption, distribution, and elimination, protecting against xenobiotic exposure, and conferring multidrug resistance in cancer, particularly acute myeloid leukemia.
