Background
MTCO1 (also called COX1, COI, or MT-CO1) is the mitochondrially encoded cytochrome c oxidase subunit I, a 57 kDa, 513-amino-acid integral membrane protein that forms the catalytic core of respiratory Complex IV (cytochrome c oxidase), the terminal enzyme of the mitochondrial electron transport chain that reduces molecular oxygen to water and pumps protons to drive ATP synthesis . Located on the heavy strand of mtDNA from nucleotide 5904–7444, MTCO1 is one of only three mtDNA-encoded subunits (together with MTCO2 and MTCO3) among the 13 that make up mammalian Complex IV . Mutations or altered expression of MTCO1 have been linked to a spectrum of disorders including Leber’s hereditary optic neuropathy, Complex IV deficiency, sideroblastic anemia, sensorineural deafness, colorectal cancer, and recurrent myoglobinuria, and its levels are modulated by regulators of mitochondrial biogenesis such as PGC-1α .
