Background
PI3 Kinase p110α (phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha) is a 1,068-amino-acid lipid kinase that, in complex with its regulatory subunit p85, phosphorylates PIP₂ to generate the second messenger PIP₃, thereby activating the PI3K/AKT/mTOR pathway that controls cell growth, survival, metabolism, and angiogenesis; encoded by PIK3CA, it contains an N-terminal adaptor-binding domain, a Ras-binding domain, a C2 domain, a helical domain, and a C-terminal kinase domain, and is the most frequently mutated oncogene in human cancers, with hotspot mutations (e.g., E545K and H1047R) causing constitutive lipid kinase activity and driving tumorigenesis.
